BOSTON (July 29, 2012) Researchers from the Massachusetts Eye and Ear Infirmary, The Children's Hospital of Philadelphia, Loyola University Chicago Health Sciences Division and their collaborators have isolated an elusive human gene that causes a common form of Leber congenital amaurosis (LCA), a relatively rare but devastating form of early-onset blindness. The new LCA gene is called NMNAT1. Finding the specific gene mutated in patients with LCA is the first step towards developing sight-saving gene therapy.
LCA is an inherited retinal degenerative disease characterized by reduced vision in infancy. Within the first few months of life, parents usually notice a lack of visual responsiveness and unusual roving eye movements known as nystagmus. LCA typically involves only vision problems, but can be accompanied by disease in other organ systems in a minority of patients. LCA is a common reason children are enrolled in schools for the blind.
"The immediate benefit of this discovery is that affected patients with mutations in this new LCA gene now know the cause of their condition," said Eric Pierce, M.D., Ph.D., co-senior author and director of the Ocular Genomics Institute at Mass. Eye and Ear. "Scientists now have another piece to the puzzle as to why some children are born with LCA and decreased vision. The long-term goal of our research is to develop therapies to limit or prevent vision loss from these disorders."
NMNAT1 is the 18th identified LCA gene. The gene resides in a region that was known to harbor an LCA gene since 2003, but the specific disease gene has been undiscovered until now. These findings will be published on July 29 in the online edition of Nature Genetics.
To identify NMNAT1, scientists performed whole exome sequencing of the family of two siblings who initially presented for evaluation of LCA but who had no mutations in any of the known LCA genes. Being seen by a multi-disciplinary team that took the case from careful clinical characterization to genetic testing to the research laboratory was an essential ingredient for success.
"By using whole exome sequencing, we found a mutation in a gene that no one could have predicted would be associated with LCA," said Dr. Pierce.
"Whereas most of the known LCA genes involve dysfunction of retinal ciliary proteins necessary for light detection in the eye, NMNAT1 is uniquely distinguished by being the first metabolic enzyme linked to LCA," said Marni J. Falk, M.D., co-first author and Clinical Geneticist at The Children's Hospital of Philadelphia and University of Pennsylvania Perelman School of Medicine.
Having found a mutation in NMNAT1 in this one family, the investigators next asked if mutations in NMNAT1 also cause disease in other patients with LCA. Screening of 284 unrelated patients with LCA from the United States, England, France and India allowed them to identify 13 other patients with mutations in NMNAT1 as the cause of their disease.
Drs. Falk, Pierce and colleagues also studied how the identified mutations in NMNAT1 affect the function of the NMNAT1 protein, and thus may cause dysfunction and death of the light sensitive photoreceptor cells in the retina. Working together with Eiko Nakamaru-Ogiso, Ph.D., in the Department of Biochemistry and Biophysics at The University of Pennsylvania, they found that mutations in NMNAT1 appear to decrease the ability of the NMNAT1 protein to produce NAD+, a key mediator of cellular signaling and energetics.
Early treatment for patients with NMNAT1-related LCA could be especially beneficial.
Researchers found that all but the youngest patient with NMNAT1 mutations had damage to the macula, the center of the retina that is needed for central vision. "This 4-year-old girl who doesn't have central vision loss yet can possibly benefit substantially if we can devise a therapy for her NMNAT1-mediated LCA that prevents her from developing severe central vision loss," Dr. Pierce said.
This study is an example of the multidisciplinary collaboration among the three institutions, using exome sequencing to discover genes involved in inherited diseases caused by mutations of a single gene. "With the robust database and pipeline that we have developed, we have analyzed more than 300 whole exomes of patients and families with single-gene diseases," said Dr. Xiaowu Gai, co-senior author and director of the Center for Biomedical Informatics at Loyola University Chicago Stritch School of Medicine. "We are following up on a number of strong candidate genes. We are sequencing many new samples and expect similar exciting discoveries for other diseases."

Some cycling activists scuffled with police officers during the Olympics opening ceremony and more than 100 people were arrested, authorities said.
The cyclists were members of a monthly cycling event called "Critical Mass" that normally passes through the area, but a police order had prohibited the group from traveling into the Olympic Park area in east London during Friday night's opening ceremony.
Dozens of cyclists were blocked by police vans from proceeding, Scotland Yard said.
Two groups of protesters were arrested on suspicion of breaching protest conditions and causing a public nuisance, Scotland Yard said.

ROCHESTER, Minn. (AP) ? Illinois Congressman Jesse Jackson Jr., whose whereabouts haven't been disclosed since he mysteriously took a medical leave several weeks ago, is being treated for depression at the Mayo Clinic in Rochester, Minn., according to a statement Friday from the hospital.
Jackson is undergoing an extensive inpatient evaluation for depression and for gastrointestinal issues, according to the statement, which also provided the first details about his medical condition. But it provided no details about where the congressman, the son of civil rights leader Jesse Jackson, had previously been staying.
"Further information will be released as Congressman Jackson's evaluation proceeds," the statement said, adding that he and his family were "grateful for the outpouring of support and prayers that have been received throughout his care."
Jackson went on leave June 10, though his office didn't disclose it until weeks later and has been mum on details ever since. Initially, his office said Jackson was being treated for exhaustion. But his staff later said Jackson's condition was more serious and required inpatient medical treatment. They also said Jackson has been grappling with emotional issues.
Under mounting pressure from fellow politicians and others, his office released a statement last month from his unnamed doctor saying Jackson was receiving intensive medical treatment at a residential treatment facility for "a mood disorder."
The timing of the leave has raised questions, in part because Jackson is facing an ethics investigation in the U.S. House connected to imprisoned former Illinois Gov. Rod Blagojevich.
The House Ethics Committee is investigating allegations that Jackson was involved in discussions about raising money for Blagojevich's campaign in exchange for the then-governor appointing him to President Barack Obama's vacated U.S. Senate seat. Blagojevich is serving a 14-year prison sentence for corruption.
Jackson was not charged and has repeatedly denied wrongdoing.
More than 3,000 members of the public as well as primary and secondary school students participated in the ?My Future Hong Kong? drawing competition organised by the Planning Department and City Gallery, and supported by the Hong Kong Society for Education in Art, to illustrate their vision of the future through painting.
The award presentation ceremony, in which 48 participants and 2 schools received awards, was held at the City Gallery in Central today (July 28).
Officiating at the ceremony, the Director of Planning, Mr Jimmy Leung, said he was encouraged by the fervent response to the competition.
?The drawing competition is intended to encourage the public to illustrate their ?dream homeland? and the future of the city using imagination and creativity, so as to share their vision of Hong Kong as the ideal place to live,? Mr Leung said.
The competition comprised three sections, namely the primary school section, secondary school section and open section, with the respective themes of ?My Dream Homeland?, ?Ideal Green City? and ?Impressive Hong Kong?. A champion and 15 outstanding awards were selected for each section. Awards for active participation were also given to the primary and secondary schools with the highest number of submissions.
The winning entries will be exhibited in the City Gallery from August to mid-September.
Located at 3 Edinburgh Place, Central, the City Gallery will be officially opened in the latter half of August. It will be the first permanent planning and infrastructure gallery in Hong Kong. Occupying five storeys with a floor area of more than 3,200 square metres, the gallery will offer a host of interactive exhibits to showcase the uniqueness of Hong Kong past and future, as well as share with the public the vision for city planning and infrastructure projects.

Oh wow, the past three weeks have been a nightmare of tiredness and nausea. No amount of sleep or ginger ale seem able to relieve either. I found the first three months of my last pregnancy extremely hard-going, much tougher than the following six months, and it seems that this one is set to be the same. I can?t get my brain or body going and am counting down the days?until the rock-star feeling of the second trimester. Please let me have that rock-star feeling again! Minus the cankles and the massive boobs would be nice though.
I find myself looking back to my first pregnancy quite a lot but my main memories of the initial three months are trying to make a quick but discreet dash to the loo to throw up every ten minutes. After narrowly missing my boss? shoes last week, I think it?s fair to say that discretion has gone out of the window already ? ?Rachel, do you have something to tell me?? Some things are just so hard to hide, aren?t they?
I nervously made my way to see my gynecologist for the first scan on Tuesday. Even after a positive pregnancy test, I found myself still unwilling to believe the result without his confirmation. But, at 6 weeks and 2 days, I saw my little 3.5mm baby and it?s ever so tiny heartbeat on the screen in front of me. It was an absolutely joyous moment and now it?s finally starting to sink in that, all things going well, we?re going to be a family of four in 2013.
Now I just need to find an extra boost of energy to see me through until the morning sickness and tiredness abates. It?s CK that I feel most sorry for as I find myself having to sit down more often rather than playing with him. But we?ve been out for longer walks which he seems to like ? I?m finding that walking at a slow but steady pace and filling my lungs with fresh, Swiss air is doing me the world of good and keeping CK entertained without me having to exert too much energy. Shame the weather has made a turn for the worse this week.
My next scan is a mere two and a half weeks away. I find it very reassuring to have them so frequently. At any point in time, I?m four weeks away at the most from knowing that something is wrong. Maybe the Swiss are a little cautious compared to the Brits but when it comes to the health of my unborn child, I?m so thankful for it. Plus, I get to build up quite a gallery even before the little one is born.
So now I must bid you farewell so I can rush off and down another glass of ginger ale and take a nap. Oh what an exciting life I lead ? but I wouldn?t have it any other way.

Kale/Spinach Smoothie: A regular in our household
WARNING: Reading the following might result in improved health, more energy, and a desire to swing from a tree branch. Oh, and your kids will benefit as well!?
This morning my husband, my mother-in-law, daughter, and I shared a simple breakfast of scrambled eggs,?saut?ed?fresh squash from the farmer?s market, and cups of coffee for the adults (no cream/sugar.) Vivi wasn?t crazy about the squash, but tasted it several times, examined the texture, and eventually decided to enjoy her eggs without. She still seemed hungry, so I quickly blended up some raw kale, frozen fruit, coconut milk, and water. She gulped down three small cups.
This meal, while simple enough, met the complex and diverse needs of all of us. The low-glycemic index of the squash and eggs satisfied my diabetic mother-in-law. My husband, who seems to be always trying to slim down and get a six-pack (I support ya, sweetie!), appreciated the low-carb aspect of the meal, but felt satiated by the coconut oil used to cook the squash. And my daughter? Well at 16 months, a meal like this is normal, tasty, and?unbeknownst?to her,?nutritious. When we eat Paleo as a family, every meal is a home run: nutritious, tasty, and satisfying.
Paleo is a total change of lifestyle and can be tough enough for adults. So often, there might be one person in a family eating Paleo, while the rest stick to what?s familiar. I want to assure you that it doesn?t have to be this way! But inevitably, there are usually two questions floating around when you mix Paleo and kiddos:
What does Paleo eating look like for babies, toddlers, and older children?
Paleo eating for kids is pretty simple: if you?re eating Paleo, they can eat all the same things as you. No more cooking two meals and loading the shopping cart up with ?kid-friendly? options!
Babies and toddlers will naturally gravitate toward a more veggies/fruit/fat based version and less meat. My daughter will often refuse meat for a week at a time, while devouring everything else. Paleo and baby-led weaning naturally go hand in hand: think soft pieces of fruit and vegetables, egg yolks, homemade broths, and slow-cooked meats that are easy to chew for the baby who is at least 6 months old. Don?t forget the breastmilk?your cavewoman ancestors would be proud!
Independent toddlers will relish the opportunity to eat what their parents do, rather than something else that looks and tastes totally different. Having options to choose from will allow these headstrong babes to feel in control.
Older children might be a bit more difficult to get on board if they haven?t eaten this way previously. The best thing you can do is to only keep nutritious options in your home, make Paleo meals, and don?t worry about the rest. Kids are so?resilient?and adaptable: give them the chance to make healthy choices, and I think you will be surprised.
And is Paleo eating even healthy for kids??
Immediate answer: YES! Do you know anyone who has ever become obese/sick/diseased while eating plenty of vegetables, fruits, and high-quality meats, eggs, fats, and fish? Children thrive on this type of diet, and you may have seen something similar prescribed for kids with Autism Spectrum Disorder, the GAPS diet. ?The Paleo way of eating provides such a varied, nutrient-rich diet that allows a parent to relax about food choices. No need to worry if your little one is getting enough of one nutrient or another because, most likely, they are getting them all and in ample?quantities.
However, there are two big issues that concern many parents: no grains and no dairy. Aren?t kids supposed to have both for the fiber and nutrients (grains) and the calcium and vitamin D (dairy)?
The short answer is this: kids will get all of the above nutrients from a Paleo diet. Vegetables and fruit provide ample fiber (yay poop!), leafy greens and nuts take care of calcium, and vitamin D can be sourced naturally from a short play session outside (step away from the Wii, my friends.) Instead of cow?s milk, my daughter drinks plenty of water, kale/spinach shakes, and an occasional cup of straight coconut milk. I?m pretty positive (at least 99% sure) that she is not only meeting the government?s snazzy RDA (recommended daily amount) of nutrients, but blowing those old-school recommendations out of the water. But if you want scientific charts, medical studies, etc.,?and not the word of some random mama on the internet, you can?t do wrong with reading a little Robb Wolf.
Kids on a Paleo?diet are healthy, happy, don?t experience sugar-induced mood swings, and will typically sleep pretty soundly. If the warm and fuzzy image of your kids growing lean and strong on whole, nutrient-dense foods and plenty of play outside has you clamoring for more info, take a gander at these oh-so-helpful resources:

Noe Valley/Glen Park: La Ciccia It may be hidden in the hinterlands straddling the sleepy 'hoods of Noe Valley and Glen Park, but La Ciccia, a cozy family-run Sardinian spot, is Grand Central on any given night. Mission: Blue Plate There's no shortage of hipster restaurants in the city, and Blue Plate has all the usual elements: PBR, adult mac 'n' cheese and tattooed staffers. Still, the low-key spot (look for the vintage EAT sign) serving up gussied-up, sustainably sourced New American comfort food, along with serious wines, is that proverbial neighborhood spot for a certain foodie demographic living on the south side of the city. Bonus points for its secret garden.

Richmond: Chapeau! When tourists want French food, they usually head Downtown to the better known addresses, but locals who live west of Van Ness know the real deal can be had at Chapeau! where gregarious chef/owner Philippe Gardelle still regales les dames with kisses on both cheeks, les monsieurs with a firm handshake and everyone with textbook bistrot classics and well-priced vins including a can't be beat early-bird and weekday prix fixe. This long-running venture used to be in an even more obscure Richmond locale, but now it's a firmament on the Clement Street drag where you can walk off your cassoulet.

Dogpatch: Piccino Most city folks are just getting word of this far-flung Dogpatch neighborhood, but the local artists, artisans and other free-minded residents who've been turning around this up-and-coming neighborhood off Third Street have long depended on Piccino?for sustenance. Even since moving to its much larger Yellow Building digs, the Cal-Italian, which turns out delightful ingredient-driven, thin-crust pizzas, salads and one helluva a meatball, gets packed to the gills most hours of the day.

North Beach: The House A welcome respite in the land of red-sauce restaurants and red-light strip clubs, The House, this low-key Asian-fusion beckons North Beach locals and anyone else tired of Italian to its tiny blonde wood digs where pioneering chef Lary Tse's turns out colorful, architectural creations like miso-marinated cod and roasted unagi, proffered by a skillful, yet friendly staff. It often gets cramped and noisy at night, but things are less hectic at lunch.

Outer Sunset: Outerlands It's truly located on the outermost edge of the city and largely dominated by surfer types in the daytime who carbo-load up on the incredibly fresh-baked bread and hearty sandwiches, but there's way more going on at Outerlands, a stealth wood-lined neighborhood haunt in the Outer Sunset where come nighttime, former Coi vet Brett Cooper crafts progressive ingredient-driven Californian dinners, which can be matched with farm-fresh cocktails. No reservations means long lines in the fog, though a scheduled winter expansion that will double the amount of seating should help alleviate some of that. In the meantime, just put your name of the list, grab a drink and deal with it. P.S. lunch is now served via table service.
Noe Valley: Firefly Most people generally don't venture this high up on Noe Valley's 24th Street unless they're heading to the park with their kid or dog, but Brad Levy's quirky eclectic neighborhood fixture Firefly has consistently been packing in the locals (and foodies who do their homework) for almost 19 years. There's plenty to lure them -- the quaint decor, the friendly service and the years-ahead-of-its-time locally sourced, global comfort menu that takes equal care of vegetarians, gluten-intolerants and omnivores alike.? Oh, and did we mention a crazy low three-course early bird prix fixe (Sunday-Thursday), the fried-chicken-of-your-dreams and Jewish holidays dinners that beat the pants of your Bubbe's?
NEW YORK (Reuters) - The euro rose against the dollar for the first time in six days on Wednesday after suggestions that European policymakers will consider new ways to tackle the region's debt crisis, while U.S. earnings supported blue-chip stocks.
Higher 2012 profit forecasts from big manufacturers Caterpillar and Boeing and an outlook for quarterly revenue growth from chipmaker Broadcom helped to offset news of an unexpected drop in new U.S. home sales.
On Wall Street, the Dow snapped a three-day losing streak and the S&P and Nasdaq slipped only modestly, even as shares of Apple Inc, the world's most valuable publicly traded company, fell 4.3 percent to $574.97 after it posted a rare miss in revenue late on Tuesday.
"I'm encouraged investors are taking the Apple news pretty well," said Jack Ablin, chief investment officer at Harris Private Bank in Chicago.
Ablin said after a more than 40 percent rise in Apple shares this year, Wednesday's drop is but a speed bump, adding that the raised outlooks from Dow components Caterpillar and Boeing "is a huge positive for the market."
The Dow Jones industrial average rose 58.73 points, or 0.47 percent, to 12,676.05. The S&P 500 Index dipped 0.42 point, or 0.03 percent, to 1,337.89. The Nasdaq Composite Index fell 8.75 points, or 0.31 percent, to 2,854.24.
The euro rose 0.8 percent to $1.2156, although the outlook remains weak and it remained only just above a two-year low of $1.2042 hit Tuesday.
A member of the European Central Bank's Governing Council, Ewald Nowotny, said there were arguments for giving Europe's new permanent rescue fund a banking license, enabling it to borrow unlimited central bank money and boosting its capacity to prevent the euro zone debt crisis from spreading.
Analysts said the market may have put too much emphasis on the comments, given other ECB officials' opposition to the idea. Investors would likely sell into the euro's rally, the analysts said.
"The market is grasping for any positive news," said Omer Esiner, chief market analyst at Commonwealth Foreign Exchange in Washington, D.C. "It adds to the already somewhat improved tone. Whether or not it lasts is the question."
Market support from central banks has been expected for weeks as economic data sags globally. Weak readings on Britain's gross domestic product and on a German business sentiment survey added to worries about slowing growth.
FED TO ACT SOON?
Top Fed officials recently spelled out what measures they might take to boost growth and hiring. Fed action could come as soon as its policy meeting next week, on July 31-August 1.
"Right now in my view it's still Fed rhetoric, but enough to keep investors confident to take risk. I don't necessarily see tangible action. I think the Fed is afraid to take action, fearful the market could shrug it off," Ablin said.
Despite a sluggish recovery and a suggestion by some analysts that the U.S. economy may already be in recession, the S&P 500 hit its highest level in 2-1/2 months last week.
The MSCI world equity index was flat on Wednesday, but has fallen 2.4 percent so far this week as concerns about the impact of Europe's problems on growth spread across the world.
U.S. dollar-denominated Nikkei futures rose 0.1 percent in choppy trading. The pan-European FTSEurofirst 300 index closed 0.07 percent lower.
Further supporting the euro, Spain and France said that a single supervisory mechanism for the bloc's banks needs to be adopted by the end of the year.
ECONOMIES SAG
The gains in the euro came despite the weak economic data from Germany and the UK, which reinforced the view that even Europe's biggest economies were being damaged by the debt crisis.
German business sentiment dropped in July for the third straight month to its lowest level in over two years, according to the latest survey by the Munich-based Ifo think tank.
British economic output also shrank much more than expected in the second quarter, hit by the euro zone debt crisis and government austerity, official data showed.
Greece was also back in the headlines with inspectors from the EU, ECB and International Monetary Fund in Athens to decide whether to keep it hooked up to a 130 billion euro lifeline or let it face default.
Three EU officials have said the team was likely to conclude Greece cannot repay what it owes, making a further debt restructuring necessary, but no decision is expected until at least September.
The benchmark 10-year U.S. Treasury note was down 3/32 in price, with the yield at 1.4008 percent after earlier rising as high as 1.44 percent. The yield hit an all-time low on Tuesday.
Gold futures rose more than 2 percent as expectations of U.S. and European monetary stimulus strengthened its appeal as an inflation hedge. Gold futures were last up 2 percent at $1,608 an ounce.
